Flow cytometry is a laser-based technology that allows for the rapid and objective analysis of cells in fluid suspension. Although peripheral blood and bone marrow specimens are the most frequently submitted samples, other body fluids (e.g., cerebrospinal, pleural, peritoneal, vitreous) or even tissues (e.g., mechanically disaggregated lymph nodes) may yield cell suspensions for analysis. Important clinical applications of flow cytometry include: lymphocyte subset analysis in congenital and acquired immunodeficiency states; classification of leukemias and lymphomas; and stem cell enumeration for transplantation.
